Title

Study of induction for cellulase production using Moringa leaf powder and Dimethylformamide (DMF) as an inducer from two different isolated Aspergillus spp. DF1 and MF5

Abstract

The study was carried out to investigate the induction of one of the most highly demanded industrial enzyme cellulase from the isolated Aspergillus spp. (DF1 and MF5) using N,N-dimethylformamide (DMF) and Moringa oleifera leaf powder as an inducer. The inductive production of enzyme was assessed by two fermentation processes i.e. Sub-merged fermentation (SmF) and Solid state fermentation (SSF). According to this study, the treatment with 1% DMF significantly increased cellulase production by approximately 60% that of medium without DMF supplementation. The maximum enzyme activity was observed i.e. 1234.6 IU/ml compared to medium without DMF supplementation i.e. 761.3 IU/ml with DF1 at 96 hours of the incubation period under SmF. Whereas, Moringa leaf powder was used as a sole source of carbon which significantly increased cellulase production by approximately 70% that of medium without Moringa leaf powder i.e. Carboxymethylcellulose (CMC). The maximum enzyme activity was observed i.e. 1378.6 IU/ml compared to medium without Moringa leaf powder i.e. 823 IU/ml with MF5 at 120 hours of the incubation period under SSF. The optimization for inductive production of cellulase using Moringa leaf powder as promising inducer compared to DMF was performed based on the modification of environmental (pH, Temperature and Incubation period) and nutritional (Nitrogen sources and Substrate concentration) parameters. By optimizing the fermentation conditions, maximum enzyme activity was recorded i.e. 1419.8 IU/ml at 120 hours of incubation period, pH 6 in SSF, 4% substrate concentration at room temperature (R.T) with Ammonium sulfate. By optimizing, Moringa leaf powder remarkably increased cellulase production by 80% that of medium without Moringa leaf powder.Then, the partial purification of enzyme was done using Ammonium Sulfate Precipitation followed by Dialysis.
